肝类器官的应用研究进展

摘 要:

肝类器官作为一种体外三维功能性组织模型,具有长期稳定传代培养和自组织的特点,并具有异质性和独特的形态学特征。在模拟肝组织结构与功能方面,肝类器官具有显著优势,在肝脏疾病发病机制研究、药物开发、再生医学以及精准医疗领域展现出巨大的潜力。其核心优势在于能够高度模拟肝脏的复杂结构与代谢功能,突破传统研究模型的局限,缩短药物研发周期。本综述主要聚焦于组织来源和诱导多能干细胞(induced pluripotent stem cells, iPSCs) 来源的肝类器官的技术进展和应用前景,并探讨当前肝类器官研究面临的机遇和挑战。

Abstract:

Hepatic organoids, as a type of three-dimensional functional tissue model in vitro, possess the capabilities of long-term stable passaging and self-organization, and exhibit heterogeneity as well as unique morphological characteristics. Hepatic organoids hold significant advantages in mimicking the structure and function of liver tissue, demonstrating great potential in the study of liver disease pathogenesis, drug development, regenerative medicine, and precision medicine. Their core strength lies in the ability to highly simulate the complex structure and metabolic functions of the liver, breaking through the limitations of traditional research models and shortening the drug development cycle. This review focuses on the technological advancements and application prospects of hepatic organoids derived from tissue sources and induced pluripotent stem cells (iPSCs), and discusses the opportunities and challenges currently faced in hepatic organoid research.
