摘 要:

CRISPR Cas12 和Cas13 蛋白反式切割活性的发现无疑是开启了“下一代分子诊断技术”的新时代。基于反式切割活性的CRISPR 诊断方法不仅在靶标核酸的检测中具有灵敏度高、特异性强、操作简便等优势,还可应用于非核酸靶标的高灵敏检测。本文总结了CRISPR 技术在临床诊断中的研究进展,内容涵盖核酸检测和非核酸检测两个应用方向。本文最后还讨论了CRISPR 技术在临床诊断应用中所面临的挑战与机遇。

Abstract:

Characterization of the trans-cleavage activities of CRISPR Cas12 and Cas13 has certainly opened the era of the next-generation molecular diagnostic technology. For the detection of target nucleic acid (NA) sequences, CRISPR has shown advantages of high sensitivity, specificity, and simplicity in operation. In addition, CRISPR-Dx can also be applied in sensitive detection of target non-nucleic acids (NNA). This review summarizes the research progress of the CRISPR technology in clinical diagnostic applications, including both NA and NNA detection. Furthermore, this review also discusses the challenges and prospects of CRISPR-Dx technology in clinical diagnostic applications.
