SIRT3在心肌代谢及相关疾病中的研究进展

摘 要:

SIRT3 是一种线粒体特异性蛋白质去乙酰化酶,在心肌组织中高表达并调控多种代谢过程。心肌作为高能量需求组织,其代谢稳态对维持心脏功能至关重要,代谢紊乱可导致心肌梗死、心肌缺血再灌注损伤、心肌肥大及糖尿病心肌病等多种心脏疾病。近年来研究表明,SIRT3 通过调控线粒体功能、氧化应激、能量代谢和细胞凋亡等多个途径,在心肌生理和病理过程中发挥关键作用。本文综述了SIRT3 的分子特征及其在心肌代谢调控中的作用机制,重点阐述了SIRT3 在各类心肌疾病发生发展中的调控网络,以期为开发以SIRT3 为靶点的心肌疾病治疗策略提供理论依据。

Abstract:

SIRT3 is a mitochondria-specific protein deacetylase that is highly expressed in myocardial tissues and regulates various metabolic processes. The myocardium, as a high-energy-demand tissue, relies heavily on metabolic homeostasis to maintain cardiac function, and metabolic dysregulation can lead to a variety of cardiac diseases, including myocardial infarction, myocardial ischemia-reperfusion injury, myocardial hypertrophy, and diabetic cardiomyopathy. Recent studies have demonstrated that SIRT3 plays a critical role in both physiological and pathological processes of the myocardium by modulating mitochondrial function, oxidative stress, energy metabolism, and apoptosis. This review summarizes the  molecular characteristics of SIRT3 and its regulation on myocardial metabolism, with a particular focus on the  regulatory networks of SIRT3 in various myocardial  diseases, aiming to provide a theoretical foundation for developing SIRT3-targeted therapeutic strategies for myocardial
diseases.
