T细胞跨血脑屏障入脑促进阿尔茨海默病的研究进展

摘 要:

阿尔茨海默病(Alzheimer’s disease, AD) 作为一种神经退行性疾病,是造成全球痴呆病例的主要原因之一,但有关AD 治疗策略的研究无突破性进展。现有研究表明T 淋巴细胞向中枢神经系统的浸润/迁移以及血脑屏障的损伤能够促进AD 的进程,但作用机制仍不完全明确。本文总结了T 淋巴细胞与血脑屏障在维持中枢神经系统稳态中的作用,介绍了AD 进程中T 淋巴细胞浸润脑实质及血脑屏障损伤的最新研究结果,分析了T 淋巴细胞突破血脑屏障浸润脑实质的作用机制和影响迁移过程的因素。综合以上内容,提出T 淋巴细胞与血脑屏障互作途径中的关键因素可以作为治疗AD 的潜在靶点,并且阻断T 淋巴细胞突破血脑屏障的关键途径或许能够成为未来AD 治疗的新策略。

Research progress on brain infiltrated T cells in Alzheimer's disease

Abstract:

As a neurodegenerative disease, Alzheimer's disease (AD) is one of the main factors for the global dementia cases. But the research on AD therapy is not encouraging. Available studies have shown that the infiltration of T cells and the damage of the blood-brain barrier can promote the process of AD, but the reasons behind these observations are still unknown. In this review, we discuss the role of T cells and the blood-brain barrier in maintaining the homeostasis of central nervous system, the latest findings of T cells infiltration to the brain and blood-brain barrier damage in AD and the mechanism and factors of T cells infiltrating the blood-brain barrier. We propose that the key factors in the interaction process between T cells and blood-brain barrier can be investigated as potential targets in AD therapy, and blocking the infiltration of T cells to the brain through the blood-brain barrier can be a potential therapeutic strategy for AD.
