中药单体逆转BRAF突变型黑色素瘤分子靶向药物耐药研究进展

摘 要:

BRAF 突变型黑色素瘤通过遗传与非遗传两种途径产生的分子靶向药物耐药性是临床治疗失败和肿瘤复发的主要原因。中医药在治疗肿瘤方面具有低毒高效、多途径、多靶点作用的优势,能为调控黑色素瘤耐药提供新的思路。研究表明,诸多中药单体可以调控BRAF 突变型黑色素瘤遗传与非遗传耐药机制,如调节胞内异常信号通路、干预细胞表型转换、抑制代谢重编程和调节表观遗传变化等,进而阻止黑色素瘤耐药的发生和发展。该文对中药单体通过调控遗传与非遗传机制抑制BRAF 突变型黑色素瘤耐药的研究现状进行总结,旨在为中药逆转黑色素瘤耐药提供理论支撑和潜在的治疗策略。

Research progress of traditional Chinese medicine monomers in reversing BRAF mutant melanoma resistance to molecular targeted therapy

Abstract:

Molecular targeted drug resistance in BRAF-mutant melanoma is a major cause of clinical treatment failure and tumor recurrence through both genetic and non-genetic pathway. Traditional Chinese medicine has the advantages of low toxicity, high efficiency, multiple-pathways and multiple-targets in the treatment of tumor, which can provide a new idea for the regulation of drug resistance of melanoma. A number of traditional Chinese medicine monomers have been shown to regulate the genetic and non-genetic resistance mechanisms of BRAF-mutant melanoma, and they can prevent the occurrence and development of drug resistance in melanoma by regulating the abnormal signal pathway, interfering with cell phenotype transition, inhibiting metabolic reprogramming and regulating epigenetic changes. Therefore, this paper will summarize the recent researches of anti-BRAF mutant melanoma drug resistance based on the traditional Chinese medicine monomer through the regulation of genetic and non-genetic mechanisms, in order to provide theoretical support and potential therapeutic strategies for traditional Chinese medicine to reverse melanoma drug resistance.
