溶瘤腺病毒靶向改造策略研究进展

摘 要:

恶性肿瘤对人类的生命健康产生了极大的威胁,生物治疗目前已经成为安全有效的恶性肿瘤治疗手段之一。溶瘤腺病毒作为一种极具潜力的生物抗肿瘤载体已引起了人们的广泛关注,其能够选择性地破坏肿瘤细胞并产生级联反应,同时释放细胞因子和肿瘤抗原激发全身抗肿瘤免疫。然而,溶瘤腺病毒在人体中的应用仍存在一些限制:预存免疫、静脉注射肝富集、靶向性不足等问题降低了病毒的感染效率和抗肿瘤活性。该文在介绍溶瘤腺病毒构建和应用进展的同时,结合本课题组的研究成果,主要对溶瘤腺病毒通过衣壳蛋白结构改造进而增强肿瘤靶向性和杀伤活性方面的研究进行归纳,以期对溶瘤腺病毒的设计和应用提供参考。

Research progress on modification strategies of targeted oncolytic adenovirus

Abstract:

Malignancy is a great threat to human lives and health. Biotherapy has already become one of the safe and effective means for treatment of malignancies. Oncolytic adenovirus has attracted extensive attention as a potential biological anti-tumor carrier, which can selectively destroy tumor cells and produce cascade reactions, simultaneously releasing cytokines and tumor antigens to stimulate systemic antitumor immunity. However, there are still some limitations in the application and practice of oncolytic adenovirus in humans, such as pre-existing immunity, liver enrichment by intravenous injection, and insufficient targeting, which reduce the infection efficiency and anti-tumor activity of the virus. In this paper, the construction and application of oncolytic adenovirus were introduced, the research on enhancing tumor targeting and killing activity of oncolytic adenovirus through structural modification of the capsid protein was summarized, combining with their research achievements, in order to provide references for the design and application of oncolytic adenovirus.
