细胞焦亡及其在肿瘤治疗中的应用潜能
摘 要:
细胞焦亡(pyroptosis) 又称细胞炎性坏死,是由消皮素(gasdermins) 介导的细胞程序性坏死,其过程中伴有炎症因子的释放。近年来,关于细胞焦亡的研究取得了较大的进展,细胞焦亡被证实与包括肿瘤在内的多种炎症相关疾病的发生发展关系密切,并展现出在肿瘤治疗中的巨大潜能。该文就细胞焦亡的生物学特点,其在肿瘤CAR-T 疗法、抗PD-1 治疗和化疗等治疗中的作用及机制进行了讨论,并重点阐述了细胞焦亡相关分子作为恶性肿瘤治疗预后和化疗方案选择的指示因子,以及作为治疗靶点协调免疫治疗、改变微环境增强疗效等方面的应用潜能。

Abstract:
Pyroptosis, also known as inflammatory necrosis of cells, is programmed cell death mediated by
gasdermins, accompanied by the release of inflammatory factors in the processs. In recent years, great progress has been made in the study of pyroptosis, which has been proved to be closely related to the occurrence and development of a variety of inflammation-related diseases, including tumors, and has shown great potential in the treatment of tumors. This review discusses the biological characteristics of pyroptosis, its role and mechanism in tumor chimeric antigen receptor (CAR) T cell therapy, anti-PD1/PDL1 therapy and chemotherapy, and emphasizes the application potential of gasdermins as indicators of malignant tumor treatment prognosis and chemotherapy regimen selection, and as a therapeutic target to coordinate immunotherapy, change the microenvironment to enhance the efficacy.
