纳米抗体在肿瘤分子影像诊断中的研究进展

摘 要:

肿瘤的分子影像诊断是以肿瘤关键分子作为识别靶点,借助特异性分子探针和高灵敏成像设备,在活体状态下对肿瘤进行成像的技术。在临床实践中因其具备可定性、定量、可视化的优点,近年来已经成为肿瘤早期诊断、肿瘤术前治疗的重要依据,在疗效监测中起到不可或缺的作用。作为可结合肿瘤特异性靶标的分子探针的重要载体,纳米抗体(nanobody, Nb) 因具备体积小、稳定性强、亲和力高、免疫原性弱等显著优势,在肿瘤分子影像诊断领域中的作用日渐突出。该文就纳米抗体的结构特征、基于纳米抗体的分子探针及其在分子影像应用领域的最新进展进行综述,同时也讨论了纳米抗体应用于肿瘤成像的临床局限性,并对其未来的研究应用进行了展望。

Abstract:

Tumor molecular imaging diagnosis is a technology that uses specific molecular probes and highly sensitive imaging equipment to image tumors in a living status, through recognizing key tumor targets. In clinical practice, tumor molecular imaging provides important evidence for early diagnosis and preoperative treatment of tumors, and plays an indispensable role in treatment effectiveness monitoring in recent years, because of its qualitative, quantitative and visual advantages. As an important carrier of molecular probes that can bind to tumorspecific targets, nanobody (Nb) has greatest strengths in the field of tumor molecular imaging diagnosis due to its small size, strong stability, high affinity, and weak immunogenicity. Therefore, Nb plays an increasingly prominent role in the field of tumor molecular imaging diagnosis. This article reviews the structural characteristics of Nb, as well as the Nb-based molecular probes and their most recent advances in molecular imaging applications. Meanwhile, this article also discusses the clinical limitations of Nb for tumor imaging, and prospects some future research applications.
