DNA损伤调节自噬调控因子1 的研究进展与展望

摘 要:

自噬是真核生物通过降解细胞内物质来维持细胞内环境稳态的一种保守机制。自噬的降解过程需要一些自噬相关蛋白和调节蛋白协调参与调节,溶酶体与自噬体融合是自噬途径降解的关键保证。其中DNA 损伤调节自噬调控因子1 (DNA damage-regulated autophagy modulator 1, DRAM1) 作为溶酶体蛋白,在自噬过程中发挥着正调控作用。DRAM1 蛋白从酵母到人中具有高度保守性,其主要存在于溶酶体上,也存在于其他细胞器,如内质网和高尔基体上。DRAM1 除调节细胞自噬,还调控细胞的凋亡,且与多种肿瘤以及感染性疾病的发生有着密切的关系。该文通过对DRAM1 在自噬以及其在肿瘤和各种疾病中的作用的最新研究进行阐述,为临床相关疾病的治疗提供潜在治疗靶点的参考。

Abstract:

Autophagy is a highly conserved intracellular mechanism in eukaryotes to maintain cell homeostasis by degrading unfunctional substances. This degradation process is coordinated and regulated by several autophagyrelated proteins and regulatory proteins. Fusion of autophagosomes with lysosomes is a key step to complete degradation in autophagy process. DNA damage-regulated autophagy modulator 1 (DRAM1) was identified as a lysosomal protein and regarded as a positive regulatory in mediating autophagy pathway. DRAM1 is highly conserved from yeast to human, which mainly distribute in lysosomes and other organelles, such as endoplasmic reticulum and Golgi apparatus. In addition, DRAM1 functions in mediating apoptotic process. Furthermore, DRAM1 involves in the occurrence of various tumors and infectious diseases. In this review, we mainly describe the most advanced research of DRAM1’s function in autophagy, additionally, its role in tumors and various diseases are introduced. Modulating DRAM1 is expected to provide a potential therapeutic targets for the treatment of clinically related diseases.
