神经酰胺激酶(CerK)催化反应的肿瘤生物学意义

摘 要:

神经酰胺激酶(ceramide kinase, CerK) 是唯一已知的在哺乳动物细胞中催化神经酰胺(ceramide, Cer) 磷酸化为1- 磷酸神经酰胺(ceramide-1-phosphate, C1P) 的脂蛋白酶。作为该酶促反应底物的Cer 在神经鞘脂代谢和信号转导途径中起着中心介质的作用,调节许多基础的细胞反应;而C1P 作为该酶促反应产物,同样是与细胞功能和命运密切相关的生物活性脂质。现有研究表明,CerK、Cer 及C1P 均参与调控肿瘤细胞增殖、存活、凋亡与迁移/ 转移,与肿瘤发生和发展密切相关,是抗肿瘤治疗的潜在靶点。

Abstract:

Ceramide kinase (CerK) is the only known lipid protein enzyme that catalyzes the phosphorylation of ceramide (Cer) to ceramide-1-phosphate (C1P) in mammalian cells. Ceramide, as the substrate of the enzyme reaction, plays a central role in sphingolipid metabolism and signal transduction pathways, regulating many basic cellular responses. C1P, as the product of the enzyme reaction, is also a bioactive lipid closely related to cell function and fate. Current researches have shown that CerK, Cer and C1P can regulate the proliferation, survival, apoptosis and migration/metastasis of tumor cells, which are closely related to the occurrence and development of tumor, and can be used as potential targets for anti-tumor therapy.
