结核分枝杆菌限制修饰系统的研究进展

摘 要:

结核分枝杆菌(Mycobacterium tuberculosis, Mtb) 是引起结核病的病原菌,其自身进化所产生的特异性和非特异性免疫机制能够抵御外来DNA ( 如噬菌体) 的入侵,其中细菌自身的限制修饰系统(restriction modification system, R-M 系统) 则是阻碍噬菌体侵染的重要原因。细菌的R-M 系统可降解噬菌体DNA,使其不能继续复制,进而不能发挥裂解细菌的作用,因此限制了结核病噬菌体疗法的应用。该文以对Mtb 基因组的生物信息学预测为基础,结合国内外相关研究,对Mtb 相关的R-M 系统研究进行综述,为探索Mtb的R-M 系统、提高结核病噬菌体疗法的疗效提供依据。

Progress of restriction modification system in Mycobacterium tuberculosis

Abstract:

Mycobacterium tuberculosis (Mtb) is a pathogenic bacterium causing tuberculosis. The specific and nonspecific immune mechanisms generated by its own evolution can resist the invasion of foreign DNA (phage etc), among which the restriction-modification system (R-M system) is an important obstacles for phage infection. Bacterial R-M systems can degrade phage DNA to prevent it from replicating and performing its role of lysing bacteria, which severely limit the use of bacteriophage therapy for tuberculosis. In this paper, based on the bioinformatics prediction of Mtb genome, and combined with domestic and abroad research, the recent advances of Mtb R-M systems were summarized, which could provide a basis for the future exploration of Mtb R-M systems and improvement of bacteriophage therapy for tuberculosis.
