磷脂酰肌醇-4-磷酸稳态调控及其在生理和病理中的作用

摘 要:

磷脂酰肌醇-4- 磷酸(phosphatidylinositol 4-phosphate, PI4P) 是真核细胞中一种含量较低,携带负电荷,分布于多种生物膜中的信号分子。PI4P 稳态对其执行多种生理功能至关重要。调控PI4P 稳态的分子机制包括影响PI4P 水平的PI4P 激酶和磷酸酶,影响PI4P 激酶活性和细胞定位的支架蛋白、小GTPase 及其翻译后修饰,以及影响PI4P 胞内分布的脂转运蛋白。该文综述了近年PI4P 的稳态调控分子机制及其在囊泡运输、脂转运代谢、细胞自噬、细胞器发生、炎性小体活化、病原菌入侵以及肿瘤发生发展等生理病理过程中的研究进展。

Homeostatic regulation of phosphatidylinositol 4-phosphate and its functions in physiology and pathology

Abstract:

Phosphatidylinositol 4-phosphate (PI4P) is an important signaling molecule with negative charge and low abundance, distributed throughout various intracellular membranes. PI4P homeostasis is necessary for its carrying out myriad cellular physiological functions. Molecular mechanisms for regulating PI4P homeostasis include phosphatidylinositol 4-phosphate kinases (PI4Ks) and its phosphatases determining intracellular PI4P levels, scaffolding proteins, small GTPases and post-translational modifications required for the activity and cellular location of PI4Ks, and some lipid transfer proteins affecting PI4P distribution within cell. This review attempts to summarize the recent progress of PI4P homeostatic regulation and functions of PI4P in membrane trafficking, lipid transfer and metabolism, autophagy, organelle biogenesis, inflammasome activation, pathogen invading, and ncogenesis and development.
