Tankyrase结构和功能的研究进展
摘 要:
Tankyrase 是一种多聚ADP 核糖聚合酶,属于PARP (poly ADP-ribose polymerase) 蛋白家族,在哺乳动物中有Tankyrase1 和Tankyrase2 两个成员。Tankyrase1 和Tankyrase2 结构相似,都包含PARP 催化结构域、SAM 结构域和ANK 结构域,Tankyrase1 的N 端还有一个功能未知的HPS 结构域,而Tankyrase2 没有。Tankyrase 参与有丝分裂、端粒维持、信号转导等多种生物学过程,与癌症、肥胖、家族性巨颌症等疾病密切相关。本文对目前Tankyrase 的结构和功能的研究进展进行了总结,以期为Tankyrase 的后续研究及其靶点药物的开发提供有意义的借鉴和参考。

Abstract:
Tankyrase, a poly ADP-ribose polymerase, belongs to the PARP (poly ADP-ribose polymerase) family and consists of two members in mammals: Tankyrase1 and Tankyrase2. Tankyrase1 and Tankyrase2 have similar structures, including the PARP catalytic domain, SAM domain, and ANK domain. Tankyrase1 contains a HPS domain with unknown function at N-terminus that is not presented in Tankyrase2. Tankyrase is involved in a variety of biological processes such as mitosis, telomere maintenance, and signal transduction, and is closely related to cancer, obesity, cherubism, and many other diseases. This paper summarizes the current research progress on the structure and function of Tankyrase, aiming to provide a meaningful reference for further research of Tankyrase and the development of Tankyrase-targeted drugs.
