抗肿瘤药物诱导肿瘤细胞副凋亡机制研究进展

摘 要:

大多数化疗药主要是通过诱导肿瘤细胞凋亡来发挥其抗癌作用。然而,癌细胞容易获得逃避凋亡的能力,对当前的化疗产生抵抗。在这种情况下,诱导癌细胞中非凋亡性细胞死亡方式以增强放化疗的敏感性、改善化疗耐药的现状,成为了新的研究热点。副凋亡(paraptosis) 是一种新的细胞程序性死亡形式,
属非凋亡性细胞死亡方式,表现为来源于内质网和线粒体肿胀的细胞质广泛的空泡化。该文综述了近年来关于副凋亡的相关背景以及抗肿瘤药物诱导副凋亡的相关分子机制,并展望了其未来的发展方向,以期为治疗癌症提供新思路和理论依据。

Advances in the mechanism of paraptosis induced by anti-tumor drugs

Abstract:

Most chemotherapeutic drugs exhibit the anti-cancer effects by inducing apoptosis in the tumor cells. However, the malignant cancer cells are easy to become resistant to the chemotherapy by obtaining the ability to escaping apoptosis. In this case, to enhance the sensitivity to chemotherapy and radiotherapy and to improve chemotherapy resistance, triggering non-apoptotic cell death in cancer cells was investigated, which becomes a new research hotspot. Paraptosis, a new type of programmed cell death, is a form of non-apoptotic cell death. It is characterized by extensive vacuolation of cytoplasm from the dilation of the endoplasmic reticulum (ER) and mitochondria. Here, we review the background of paraptosis, and the molecular mechanism of paraptosis induced by the anti-tumor drugs, and look forward the future studies of paraptosis to provide new ideas and theoretical basis for the treatment of cancer by inducing paraptosis.
