转录因子FOXO1在肿瘤形成中的调控作用及机制研究进展

摘 要:

FOXO1 (forkhead box O1) 是FOXO 家族重要成员,作为一种转录调控因子,其缺失可扰乱细胞增殖与死亡平衡,与肿瘤形成有着密切关系。FOXO1 可通过磷酸化、乙酰化、泛素化和甲基化等多种翻译后修饰方式,与PI3K/Akt、Wnt/β-catenin 等信号通路相互作用,参与调控肿瘤细胞的增殖、凋亡、EMT、氧化应激、自噬及物质代谢等多种生物学活动。该文就FOXO1 相关分子通路及其在肿瘤形成中的调控作用进行综述,探讨其作为肿瘤治疗分子靶标的可能性。

Abstract:

FOXO1 (forkhead box O1), as a transcription regulator, is an important member of FOXO family. Its deficiency can break the balance between cell proliferation and death, which is intimatly related to tumorigenesis. FOXO1 can interact with PI3K/Akt, Wnt/β-catenin or other signaling pathways through various post-translational modifications, such as phosphorylation, acetylation, ubiquitination and methylation, to participate in the regulation of proliferation, apoptosis, EMT, oxidative stress, autophagy and metabolism of tumor cells. In this review, FOXO1-related molecular pathways and its role in tumorigenesis are described, and the prospect of FOXO1 as a candidate molecular target for tumor therapy is discussed.
