Frizzled-7靶向治疗的抗肿瘤研究进展
摘 要:
Wnt 信号通路在物种进化过程中高度保守,并在肿瘤的发生发展中发挥至关重要的作用。Frizzled(Fzd) 蛋白是Wnt 受体家族,该家族中有10 个成员蛋白,Frizzled-7 (Fzd7) 为成员之一。研究表明,Fzd7 在多种恶性肿瘤中被显著上调,通过激活Wnt 通路调控细胞增殖、肿瘤侵袭与迁移,并影响肿瘤细胞干性。Fzd7 的上调与肿瘤化疗耐受以及患者的低生存率显著相关,靶向Fzd7 在体内外均表现出了良好的抗肿瘤效果。因此,Fzd7 是恶性肿瘤潜在的治疗靶点。该文对Fzd7 在癌症中的表达与功能以及Fzd7 的靶向治疗作一综述。

Abstract:
Wnt signaling pathway is highly conserved throughout evolution and plays a critical role in cancer development and progression. Wnt receptor family contains 10 members, and Frizzled-7(Fzd7) is one of key receptors. It has been demonstrated that Fzd7 is up-regulated in multiple malignancies, and involved in the proliferation, invasion, migration of tumor cells via activating Wnt signaling pathway. In addition, Fzd7 plays a key role in stem cell biology. The aberrant up-regulation of Fzd7 is significantly associated with the chemotherapy tolerance and inferior survival rates of tumor patients. Furthermore, targeted therapy of Fzd7 exhibits potent anticancer activity in vitro and in vivo. In conclusion, Fzd7 may serve as a potential therapeutic target in cancer treatment. In this article, we summarized the expression and function of Fzd7 and reviewed Fzd7-targeting therapy.
