YAP/TAZ的调控机制研究进展
摘 要:
YAP/TAZ是Hippo通路的关键效应因子,在介导Hippo通路调节器官大小及组织恶变中起着重要作用。近年来的研究发现,除了经典Hippo通路,机械信号如Rho激酶和细胞核内因素,可溶性因子如GPCR、Wnt和转化生长因子-β (TGF-β)信号通路,以及能量代谢等对YAP/TAZ也起到了多方面的调控作用。越来越多的研究表明,YAP/TAZ在肿瘤、心血管疾病和肥胖等多种疾病中发挥着重要作用。因此,深入探究YAP/TAZ的调控机制显得尤为重要。现对哺乳动物系统YAP/TAZ的调控机制研究进行综述,希望为多种疾病治疗提供新的思路。

Abstract:
Hippo pathway is a very conservative pathway in evolution and function. YAP/TAZ is a key effector of the Hippo pathway, which plays an important role in regulating organ size and tissue malignancy. Recent studies have found that in addition to the classic Hippo pathway, mechanical signaling such as Rho kinase and nuclear factors, soluable factors such as GPCR, Wnt and transforming growth factor beta (TGF-β) signaling pathway, energy metabolism, etc. also play a multi-faceted regulatory role on YAP/TAZ. As an increasing number of reports uncover the fact that YAP/TAZ plays an important role in the occurrence and development of tumors, cardiovascular diseases, obesity and other diseases, it is particularly important to explore the regulatory mechanism of YAP/TAZ in depth. This article reviews the regulatory mechanism of YAP/TAZ in the mammalian system to provide new ideas for the treatment of various diseases.
