摘 要:

甲状腺转录因子-1 (TTF-1/NKx2.1)是NKx2组织特异性转录因子家族成员之一,表达在内胚层起源的甲状腺滤泡、甲状旁腺、肺泡上皮及外胚层起源的间脑等少数组织器官中,参与上述器官的分化发育和功能维持。近来的研究显示,TTF-1的异常表达与多种人类疾病发生密切相关,且可作为相关疾病诊治的潜在新靶标。该文综述了TTF-1 的结构、表达调控、生物学功能及其在人类相关临床疾病发生发展中的作用,并展望了今后研究的方向,以期加强对TTF-1的系统了解,促进相关研究的进展,最终有助于临床疾病发生机制认识和防治新策略的开发。

Abstract:

Thyroid transcription factor-1 (TTF-1/Nkx2.1) is a member of NKx2 tissue-specific transcription factor family, which is expressed in thyroid follicle, parathyroid gland, alveolar epithelium and diencephalon originated from ectoderm, and participates in the differentiation and function maintenance of these organs. Recent studies have shown that the abnormal expression of TTF-1 is closely related to the occurrence of a variety of human diseases and can be used as a potential new target for the diagnosis and treatment of related diseases. The structure, expression regulation, biological functions of TTF-1 and its role in the occurrence and development of human-related clinical diseases were reviewed in this article in order to strengthen the systematic understanding of TTF-1 and promote the progress of related research. The future research direction of TTF-1 was also prospected. We hope these could ultimately contribute to the understanding of the pathogenesis of clinical diseases and the development of new prevention and treatment strategies.
