血管非炎性分子1的生理及病理生理作用

摘 要:

血管非炎性分子1 (vascular non-inflammatory molecule 1, vanin1; VNN1)是具有泛酰巯基乙胺酶活性的糖基化磷脂酰肌醇锚定蛋白,在人体广泛分布。VNN1催化D-泛酰巯基乙胺水解产生D-泛酸和半胱胺,前者是体内合成辅酶A的前体物质,后者则是体内具有高亲和力的抗氧化物质,它们参与体内多种生物学过程,如氧化应激、炎症反应、代谢活动。鉴于VNN1的酶学特征,它在许多疾病的发生发展中起着重要作用。该文对VNN1基因家族、VNN1蛋白结构、生物学作用及其病理生理功能进行综述。

The physiological and pathophysiological role of vanin1

Abstract:

Vascular non-inflammatory molecule 1 (vanin1, VNN1) is a glycosylphosphatidylinositol-anchored protein with pantetheinase activity, which is widely expressed in the human bodies. VNN1 catalyzes the hydrolysis of pantetheine to pantothenic acid and cysteamine, in which pantothenic acid is the precursor of coenzyme A while cysteamine is an antioxidant with high-affinity in vivo. Functionally, VNN1 is involved in many important biological processes, such as oxidative stress, inflammation and metabolism. Given the enzymatic properties, VNN1 has been implicated in the progression of many diseases. In the review, we discuss the isotype and structure of VNN1 as well as its role under physiological and pathophysiological conditions.
