高通量单细胞转录组测序发展与展望

摘 要:

近年来,单细胞转录组测序技术发展迅猛,为人们研究细胞的异质性及在单个细胞分辨率下理解生命体系、过程和不同疾病状态提供了有效手段。基于微流控技术发展而来的高通量单细胞转录组测序技术是实现大量细胞平行检测的主要方法。现从高通量单细胞转录组检测平台研究进展、单个细胞捕获及检测的常用方法——特别是微滴与微孔技术原理与特点、数据分析流程、数据挖掘手段、现有存在问题和未来发展方向等角度进行阐述,希望该技术对生命科学研究起到促进和推动作用。

Development and prospect of high-throughput single-cell transcriptome sequencing

Abstract:

In recent years, the single-cell RNA-Seq technologies have been developed rapidly. The development of the technologies provides us a power tool to understand the life processes and diseases from a cellular perspective. The high-throughput single-cell transcriptome sequencing technology based on microfluidic technology is the main method to analyze a large number of cells parallelly. In this review, we summarize the research development of high-throughput single-cell transcriptome detection platform, common methods for single cell capture and detection, especially the characteristics of microdroplet and micropore technology. We also discuss the data analysis process, data mining, current existing problems and future directions. We hope it can provide a guidance for further promotion and development of life science research.
