摘 要:

TRAF6 (tumor necrosis factor receptor-associated factor 6) 是肿瘤坏死因子受体家族中研究最为广泛且可以进行信号转导的成员之一。TRAF6 的环指结构域(ring finger domain, RF) 和锌指结构域(zinc finger motif, ZF) 对其自身泛素化活性至关重要,且它的泛素化活性受到多种因素的调节,并在PI3K/AKT、NF-κB、 Wnt 信号通路以及自噬中发挥重要作用,最终影响炎症反应、神经性疾病和癌症的发生。TRAF6 敲除的小鼠明显对疾病易感并且生存周期缩短。该文重点综述了TRAF6 的泛素化调控功能和TRAF6 自身泛素化的调控机制及其在疾病中的研究进展。

TRAF6(tumor necrosis factor receptor-associated factor 6), a member of the tumor necrosis factor receptor family, is one of the most widely studied members that can conduct signal transduction. Both the ring finger domain (RF) and zinc finger motif (ZF) are essential for the autoubiquitination of TRAF6. Moreover, the ubiquitination activity of TRAF6 is regulated by various factors and plays an important role in PI3K/AKT, NF-κB, Wnt signaling pathways, autophagy and ultimately affects inflammatory response and the development of neurological diseases and cancers. TRAF6 knockout in mice has been shown to promote the development of the disease and significantly shorten the survival time. This paper mainly reviews the regulatory mechanism and functions of TRAF6 ubiquitination and autoubiquitination in certain diseases.
