METTL3及其在肿瘤中作用的研究进展

摘 要:

甲基转移酶3 (methyltransferase-like 3, METTL3) 是m6A 甲基转移酶复合体中的关键蛋白,能够催化RNA 上的腺嘌呤(A) 第6 位氮原子发生甲基化。METTL3 介导的m6A 修饰与RNA 的剪接、出核、降解和翻译都有着密切联系,并且在肿瘤的发生发展中起着重要作用。该文主要介绍了METTL3 的结构,以及其对mRNA 翻译的调控,与非编码RNA 的相互作用等多种生物学功能在肿瘤发生发展中作用,旨在为基础及临床医学中对m6A 修饰的研究提供新的研究思路和治疗靶点。

Research progress on METTL3 and its role in tumors

Abstract:

Methyltransferase-like 3 (METTL3) contributes directly to the chemical modification of mRNA in
eukaryotes by catalyzing methylation of adenine’s sixth nitrogen atom on RNA. METTL3-mediated m6A modification exerts critical influences on RNA splicing, nuclear export, degradation and translation. Moreover,it also plays an important role in tumorigenesis. In this review, we discuss the structure of METTL3, and how its biological functions such as regulating mRNA translation and its interaction with non-coding RNA contribute to tumorigenesis and tumor progression, attempting to provide new ideas and therapeutic targets for further in-depth
research of m6A in basic and clinical medicine.
