脂质代谢调节肿瘤细胞铁死亡研究进展

摘 要:

铁死亡是一类以铁离子依赖性、脂质过氧化、线粒体退化、膜密度增加为主要特点的细胞死亡。相较于正常细胞,铁死亡更易发生于铁离子浓度高、脂质代谢异常的肿瘤细胞中,因此铁死亡相关药物对于人淋巴瘤、肾癌、纤维肉瘤等肿瘤具有较好的治疗效果。现简要介绍铁死亡相关发生机制,详细阐述脂质代谢相关基因对铁死亡的调控机理,以期加深对铁死亡以及脂质代谢的认识,为靶向铁死亡治疗肿瘤打下理论基础。

Lipid metabolism regulates ferroptosis in tumor

Abstract:

Ferroptosis is a form of regulated cell death characterized by iron ion dependence, lipid peroxidation, mitochondria shrink and membrane density enhancement. Tumor cells, compared with normal human cells, are more likely to occur ferroptosis because of high iron ion concentration and abnormal lipid metabolism. Some studies have demonstrated that targeting ferroptosis-related gene shows promising therapeutic effects in treating lymphoma, renal cancer and fibrosarcoma. This article reviews the mechanisms underlying ferroptosis, highlights
connections to lipid metabolism, helps us to get deep insights into ferroptosis and lipid metabolism, and lays the foundation for developing ferroptosis-targeted therapy.
