Furin在神经精神疾病中作用机制的研究进展

摘 要:弗林蛋白酶(Furin) 作为细胞内具有剪切活性的蛋白酶,在众多蛋白质的生产和分泌过程中通过剪切不具备生物学活性前体蛋白的特定位点,使其活化获得相应生物学功能。作为一种重要的前体蛋白转化酶,Furin 在神经系统中的作用底物主要包括一些生长因子、基质金属蛋白酶、肽类激素及其前体等,这些底物对癫痫、阿尔茨海默病、缺血性脑卒中和精神分裂症等重大神经精神疾病的发生发展具有重要的调控作用。该文概述了Furin 的基本结构与生物学功能,Furin 在一些重要神经精神疾病中的具体作用、相应机制,以及Furin 活性调节剂等方面的最近研究进展。

Abstract:

Abstract: Furin is an intracellular endoprotease that can cleave and activate many biologically inactive precursor proteins at specific sites before their secretion to exert corresponding biological functions. As an important precursor
    protein convertase, Furin has many substrates in the nervous system including growth factors, matrix metalloproteinases, as well as peptide hormones and their precursors. These substrates then play distinct important roles individually in the pathogenesis of multiple devastating brain disorders such as epilepsy, Alzheimer's disease, ischemic stroke and schizophrenia, and so on. Here we briefly introduce the basic structure and biological functions of Furin, recent research progress on its role, proposed mechanisms and activity regulator in some severe neurological
    diseases.
