β-分泌酶抑制剂研究进展

摘 要:摘 要:阿尔兹海默病(Alzheimer's disease, AD) 是一种退行性的神经性疾病,其主要病理特征为β-淀粉样蛋白(β-amyloid protein, Aβ) 的异常聚集及tau 蛋白的过度磷酸化。研究发现,β-分泌酶上调在Aβ 沉积发生发展过程中具有重要作用。抑制β-分泌酶活性有可能减少淀粉样病变,从而延缓AD 病理发生。现结合AD 的发病机制对β-分泌酶的特点及β-分泌酶抑制剂的研究进展进行综述。

Research advances in design and development of β-secretase inhibitors

Abstract: Abstract: Alzheimer's disease (AD) is a neurodegenerative disease characterized by abnormal aggregation of β-amyloid (Aβ) and hyperphosphorylation of tau protein. It has been found that the up-regulation of β-secretase plays an important role in the development of Aβ deposition. Inhibition of β-secretase activity may reduce amyloidosis, thus delaying the occurrence of AD pathology. This paper reviewed the characters of β-secretase and research advances of β-secretase inhibitors combined with the pathogenesis of AD.
