血脑屏障体外模型研发

摘 要:摘 要:血脑屏障(blood-brain barrier, BBB) 是介于血液和脑组织间的一种动态界面,严格调控血- 脑间的物质运输,为神经功能的实现提供稳定的内环境。许多药物因无法通过血脑屏障达到有效剂量而被淘汰。现在,越来越多的研究转向血脑屏障体外模型的研制。现从血脑屏障的结构、制备血脑屏障体外模型的预期目标、血脑屏障体外模型构成组分的来源以及血脑屏障体外模型装置等方面,综述了现有静态及动态培养BBB 体外模型的发展及优缺点,以期为BBB 体外模型的应用提供一定理论依据。

Abstract: Abstract: The blood-brain barrier (BBB) can be characterized as a dynamic barrier in the interface between the blood of the brain microvasculature and the brain tissue, which maintains a homeostasis that allows neurons to function properly by tightly controlling the passage of materials transport. Many therapeutic drugs with low permeability have been eliminated because they fail to cross BBB. Nowadays, more and more researches focus on the development of the in vitro BBB models. Herein, we provide a detailed review on BBB structure, the expected target of in vitro model, the components used to form the BBB model, and the advantages and disadvantages of in vitro model device, thereby intending to contribute to a stronger theoretical basis for the application of in vitro BBB model.
