Calsyntenins家族在学习记忆和阿尔茨海默病发病中的作用研究

摘 要:摘 要:钙结合蛋白家族(calsyntenins, CLSTNs) 属于细胞黏附分子钙黏素超家族,包括CLSTN1、CLSTN2和 CLSTN3 三个成员,主要表达于锥体神经元突触后膜和转运囊泡中,参与调控线虫和人的学习记忆。CLSTNs 能将细胞外水解信号和细胞内Ca2+ 激活信号相关联,还是一个调控多个膜结合细胞器转运的胞内运输蛋白。现分析CLSTNs 激活 Ca2+ 信号途径和参与胞内转运的功能,归纳CLSTNs 调控学习记忆、突触发育和突触传递的作用,以及在阿尔茨海默病(Alzheimer’s disease, AD) 发病中的作用,为理解CLSTNs 的细胞生物学功能提供线索。

Research on calsyntenins family and its role in learning-memory and pathogenesis of Alzheimer’s disease

Abstract: Abstract: The calsyntenins (CLSTNs) are atypical cadherins members of the cadherin superfamily comprising calsyntenin 1, calsyntenin 2 and calsyntenin 3. These proteins are primarily expressed in the postsynaptic membranes and transport vesicles in pyramidal neurons. They have been shown to play a role in memory in both Caenorhabditis elegans and human. CLSTNs can mediate cell-cell adhesion with the unique potential to link extracellular proteolytic activity with intracellular Ca2+ signaling. In addition, CLSTNs are also a group of transmembrane cargo-docking proteins for kinesin-1-mediated vesicular transport. In order to understand the functions of CLSTNs in cell biology, this review delineates CLSTNs function in the activation of intracellular Ca2+ signaling and in vesicular transport, and most importantly, summarizes the roles that involved in regulation of learning-memory, synapse formation, and in the pathogenesis of Alzheimer’s disease.
