去泛素化酶USP14的功能及其抑制剂的研究进展

摘 要:摘 要:去泛素化酶USP14 (ubiquitin-specific proteases 14) 属于去泛素化酶(deubiquitinating enzymes, DUBs)中的泛素特异性蛋白酶家族(ubiquitin-specific proteases, USPs),其可以切除底物的泛素链,并发挥相应的生物学效应。USP14 在信号通路转导、病毒感染的调节、神经系统功能以及肿瘤发生发展进程中均起着重要作用。因其在肿瘤发展中的重要作用以及作为药靶的可行性,当前已经有很多研究将USP14 的抑制剂作为抗肿瘤药物研究的新方向。主要综述了USP14 在信号通路、病毒感染、神经系统、肿瘤发展中的作用以及其抑制剂的研究进展。

Abstract: Abstract: Ubiquitin-specific protease 14 (USP14) is one of the ubiquitin-specific proteases (USP) family of deubiquitinating enzymes (DUBs), which could exert biological effects via trimming ubiquitin from its substrates. USP14 plays an important role in cell signaling transduction, regulation of viral infection, neurological function and carcinogenesis. Many researchers regard USP14 inhibitor as a novel anti-tumor therapy strategy, because USP14 is feasible to be a drug target and plays an important role in tumor progression. In this review, we discuss the research advance of USP14 in signaling pathway, viral infection, nervous system and cancer development, as well as the latest study of USP14 inhibitors.
