均相光激化学发光免疫分析技术研究进展

摘 要:摘 要:均相光激化学发光免疫分析技术(amplified luminescent proximity homogeneous assay linked immunosorbent assay, AlphaLISA) 是一种以表面包被有亲和涂层的受体微球(acceptor beads) 和供体微球(donor beads) 为载体的均相免疫学检测技术。该技术灵敏度高、特异性强、背景影响低、样品要求低、耗费少、分析操作简单以及检测范围宽泛,既适用于传统ELISA 和其他化学发光技术可以检测的小分子简单相互作用,更可以胜任大分子复杂相互作用的高通量检测。主要针对AlphaLISA 技术的特点和较之其他免疫学检测技术的优势进行总结,并对其在生物标记物(biomarker) 检测及疾病临床检验方面的应用进行综述。

Abstract: Abstract: Amplified luminescent proximity homogeneous assay linked immunosorbent assay (AlphaLISA) is a kind of homogeneous immunoassay based on affinity-coated silica gel beads. It’s a technique of high sensitivity and specificity, low background interference, low sample cost, simple operation and wide detection range. AlphaLISA iscapable for detecting both small molecule interactions which can also be detected by conventional ELISA and other chemiluminescent techniques, and large or more complex molecules interactions in high-throughput. Here we briefly summarized the characteristics of AlphaLISA and its advantages comparing to other immunological screening techniques, and reviewed its application in biomarker detection and clinical diagnosis of disease.
