低氧诱导因子-1α的翻译后化学修饰及其对活性的调控作用

摘 要:摘 要:低氧诱导因子-1(hypoxia inducible factor-1, HIF-1)是参与调节机体氧平衡的重要转录因子。作为一种蛋白质,它的主要亚基HIF-1a受到多种翻译后的化学修饰,如泛素化、羟基化、乙酰化、类泛素化、磷酸化、巯基化等的影响,从而使其蛋白的稳定性、入核以及对下游基因的促转录活性受到调节。因此,研究HIF-1α的翻译后修饰,不仅有助于我们加深对HIF-1作为一个多种疾病的治疗靶标的认识,更能为我们理解类似转录因子自身的调控提供思路。

The post-translational chemical modification and its regulating effects on the activity of hypoxia inducible factor-1α

Abstract:
Abstract: Hypoxia inducible factor-1( HIF-1 ) is an important transcription factor in oxygen homeostasis of the organism. As a protein, its major subunit HIF-1a is subjected to a variety of the post-translational chemical modification, such as ubiquitination, hydroxylation, acetylation, sumolyation, phosphorylation, sulfhydrylation, etc. By these modifications, the stability, cytoplasmic-nuclear trafficing and transcriptional activity of HIF-1a are regulated. Therefore studies in this aspect are helpful in understanding not only HIF- as a therapeutic target for the diseases but also the regulation of other similar transcriptional factors.
