摘 要:摘 要:核苷三磷酸焦磷酸酶(nucleotide triphosphates pyrophosphatase, NTP-PPase) 是一类可将核苷三磷酸(nucleotide triphosphates, NTPs) 水解生成其对应的NMPs,同时释放出焦磷酸的核酸酶。随着其在从大肠杆菌到哺乳动物细胞中的广泛发现,它们在进化上的保守性表明该家族分子在维持生命过程中具有重要作用。NTP 焦磷酸酶可以通过水解异常核苷酸,避免其掺入到新合成的DNA 或RNA 链中,保证DNA 合成的精确性和基因组的稳定性,并与肿瘤的发生、发展密切相关。将对NTP 焦磷酸酶的研究进展作简要介绍。
Abstract: Abstract: NTP pyrophosphatases are capable of hydrolyzing NTPs into di- or monophosphates. With the prevalent presence from Escherichia coli to mammalians, its evolutional conservation suggests the critical roles in maintaining proper life processes. NTP pyrophosphatases guarantee the fidelity of DNA replication and genome stability through hydrolyzing non-canonical NTPs to avoid the incorporation of these non-canonical nucleotides into daughter DNA. They are demonstrated to be related to tumorigenesis as well as chemotherapy drug resistance. In this review, we intend to summarize the recent research progresses in NTP pyrophosphatases.
