高通量siRNA筛选技术发展与展望

摘 要:RNA 干扰(RNA interference, RNAi) 技术被认为是20 世纪生命科学领域最伟大的发现之一,是最具发展前景的生物医药技术和基因功能研究方法。因此,高通量siRNA 筛选技术也成为生物学最强有力的研究工具之一。特别是在当今后基因组时代,高通量siRNA 筛选技术已经成为揭示复杂信号网络调控、寻找新药物靶点分子等诸多问题的利器。将对这一技术的产生和发展进行回顾,总结描述我们的筛选经验,通过介绍若干典型筛选工作,来梳理高通量筛选技术的发展脉络,最后展望了高通量siRNA 筛选技术的发展趋势。

Development and perspective of high-throughput siRNA screening technology

Abstract: RNA interference (RNAi) has been considered as one of the most important discoveries in 20th century. It has been widely applied in drug discovery and genetic network research, thus high-throughput screening (HTS) having become the most powerful tool. Especially in post-genome period, HTS has been applied to unveil the regulatory network, to identify new drug target, and so on. In this work, we will go over the history of HTS, describe our experience, illustrate the typical works, and finally discuss the perspective of HTS.
