单核细胞趋化蛋白诱导蛋白-1的研究进展

摘 要:摘 要: 单核细胞趋化蛋白诱导蛋白-1 (MCPIP1) 是最近发现的一类具有免疫调节作用的CCCH 型锌指家族分子。MCPIP1 可被LPS、IL-1β 或MCP-1 等多种炎性因子刺激表达,可通过下调炎症因子( 如IL-6、IL-12p40 等) 表达,从而负向调控炎症过程。MCPIP1 的作用机制主要是作为RNA 酶调节某些炎性因子mRNA 或pre-miRNA 的降解。此外,MCPIP1 也可作为去泛素化酶靶向TNF 受体相关蛋白(TRAFs) 成员,从而负向调控JNK 和NF-κB 信号活化。将从MCPIP1 分子的发现、基因和蛋白质结构、生物学功能、表达调控以及临床应用前景等几个方面进行阐述。

Abstract: Abstract: MCP-1-induced protein-1 (MCPIP1) is a newly identified CCCH zinc finger protein that is induced in human monocyte-derived macrophages when stimulated by monocyte chemotactic protein (MCP-1) or IL-1β. MCPIP1 plays a key role in the down-regulation of inflammatory cytokines such as IL-6 and IL-12p40. The underlying mechanism that MCPIP1 negatively regulates inflammatory factors has been studied at different levels. MCPIP1 is an essential RNase and down-regulates specific mRNAs of cytokines via a conserved region in the non-AU-rich element of the 3′-UTR. Furthermore, MCPIP1 acts as a deubiquitinase to negatively regulate JNK and NF-κB signaling by targeting the TNF receptor-associated factors (TRAFs). In this review, we summarized recent research progress and discussed the structure, biological function, regulation and mechanism of MCPIP1, as well as its clinical application potential.
