摘 要:摘 要: 蛋白质相互作用在细胞凋亡与坏死、新陈代谢及生长繁殖等过程中具有重要作用。蛋白质相互作用研究技术包括酵母双杂交、串联亲和纯化、GST pull down、免疫共沉淀、蛋白质芯片、双分子荧光互补技术、Far-western 印迹等。另外,生物信息学方法在蛋白质相互作用研究中也得到广泛运用,建立了大量的蛋白质相互作用数据库。蛋白质相互作用异常将导致许多疾病,抑制这些相互作用为治疗疾病提供了一种创新性方法。
Abstract: Abstract: Protein-protein interactions play important roles in the apoptosis, necrosis, metabolism, growth and so on. Here we summarized the research techniques in protein-protein interactions, including yeast two-hybrid system, tandem affinity purification, GST pull-down, co-immunoprecipitation, protein chip, bimolecular fluorescence complementation and far-western blotting. Bioinformatics has been used widely in the study of protein-protein interactions, and a number of databases have been constructed. The abnormality in protein-protein interactions may be associated with human diseases. The inhibition of these abnormal interactions will provide an innovative therapy in future.
