低氧诱导因子-1激活的调节

摘 要:摘 要:低氧诱导因子-1(hypoxia-inducible factor-1, HIF-1)是细胞对缺氧反应的关键调节者,能激活100多种与缺氧条件下细胞存活有关的基因转录。它是由α和β两个亚基组成的异源二聚体,而HIF-1α是决定其活性的功能性亚基。HIF-1α的合成主要受PI3K及MAPK通路调节。在常氧条件下,由于氧依赖性的及氧非依赖性的通路HIF-1α被降解,几乎没有活性。当细胞缺氧时,HIF-1α积累,然后通过输入蛋白的转运进入细胞核,与HIF-1β形成HIF-1,后者与辅激活因子一起与靶定DNA结合,从而发挥其转录激活功能。就HIF-1激活的调节作一综述。

The regulation of HIF-1 activation

Abstract: Abstract: Hypoxia-inducible factor-1 is a key regulator of the cellular response to hypoxia, activating the transcription of more than 100 genes crucial for adaptation to hypoxia. It is a heterodimer of the regulated HIF-1α subunit and the constitutively expressed HIF-1β subunit. The synthesis of HIF-1α is mainly regulated by the PI3K and MAPK pathways. It is degraded through oxygen-dependent or oxygen-independent mechanisms under normoxic condition. Under hypoxic condition, HIF-1α accumulates, translocates into the nucleus by importins, and interacts with HIF-1β to form HIF-1. Together with coactivator, HIF-1 binds the targeted-DNA to exert the transcriptional activation function. Here we reviewed the regulation of HIF-1 activation.
