摘 要:摘 要:Nicastrin(NCT)是高度糖基化的I型跨膜蛋白,是g-分泌酶复合物的重要组件蛋白之一,广泛分布于人类或鼠的所有细胞类型。它不仅与g-分泌酶的组装和成熟密切相关,其构象及表达变化对g-分泌酶活性和阿尔茨海默病(Alzheimer抯 disease, AD)中b淀粉样蛋白(amyloid protein b, Ab)的产生及降解也起重要调节作用。本文就国际上近几年在NCT的结构、合成、分布、降解及功能等方面的研究进展作一综述。

Abstract: Abstract: Nicastrin(NCT), one of the most important components of g-secretase, is a highly glycosylated type I transmembrane glycoprotein. It is expressed in almost all cell types in human and mouse. NCT is not only closely related to the assembly and maturation of g-secretase complex, more importantly, changes of its conformation and expression have a significant effect on the regulation of g-secretase activity, as well as the generation and degradation of amyloid b peptide in Alzheimer抯 disease. This review summarizes the development of structure, synthesis, distribution, degradation and function of NCT in recent years, including that in our laboratory.
